Cardiovascular Overexpression of Transforming Growth Factor-b1 Causes Abnormal Yolk Sac Vasculogenesis and Early Embryonic Death

چکیده
Transforming growth factor-b1 (TGF-b1) is expressed in the adult and embryonic vasculature; however, the biological consequences of increased vascular TGF-b1 expression remain controversial. To establish an experimental setting for investigating the role of increased TGF-b1 in vascular development and disease, we generated transgenic mice in which a cDNA encoding a constitutively active form of TGF-b1 is expressed from the SM22a promoter. This promoter fragment directs transgene expression to smooth muscle cells of large arteries in late-term embryos and postnatal mice. We confirmed the anticipated pattern of SM22a-directed transgene expression (heart, somites, and vasculature of the embryo and yolk sac) in embryos carrying an SM22a–b-galactosidase transgene. SM22a– b-galactosidase transgenic mice were born at the expected frequency (13%); however, nearly all SM22a–TGF-b1 transgenic mice died before E11.5. SM22a–TGF-b1 transgenic embryos identified at E8.5 to E10.5 had growth retardation and both gross and microscopic abnormalities of the yolk sac vasculature. Overexpression of TGF-b1 from the SM22a promoter is lethal at E8.5 to E10.5, most likely because of yolk sac insufficiency. Investigation of the consequences of increased vascular TGF-b1 expression in adults may require a conditional transgenic approach. Moreover, because the SM22a promoter drives transgene expression in the yolk sac vasculature at a time when embryonic survival is dependent on yolk sac function, use of the SM22a promoter to drive expression of “vasculoactive” transgenes may be particularly likely to cause embryonic death. متن کاملCompensatory signalling induced in the yolk sac vasculature by deletion of TGFbeta receptors in mice.
Vascular development depends on transforming growth factor beta (TGFbeta), but whether signalling of this protein is required for the development of endothelial cells (ECs), vascular smooth muscle cells (VSMCs) or both is unclear. To address this, we selectively deleted the type I (ALK5, TGFBR1) and type II (TbetaRII, TGFBR2) receptors in mice. Absence of either receptor in ECs resulted in vasc...
متن کاملTransforming growth factor-beta signalling in extraembryonic mesoderm is required for yolk sac vasculogenesis in mice.
We have analysed the function of transforming growth factor beta (TGF-beta) in yolk sac development in mice by generating somatic chimaeras in which the extraembryonic mesoderm, which gives rise to the endothelial and haematopoietic cells of the yolk sac vasculature, is derived from embryonic stem (ES) cells. The ES cells were stably transfected and express either the full-length type II bindin...
متن کاملRole of mutation and pharmacologic block of human KCNH2 in vasculogenesis and fetal mortality: partial rescue by transforming growth factor-β.
BACKGROUND N629D KCNH2 is a human missense long-QT2 mutation. Previously, we reported that the N629D/N629D mutation embryos disrupted cardiac looping, right ventricle development, and ablated IKr activity at E9.5. The present study evaluates the role of KCNH2 in vasculogenesis. METHODS AND RESULTS N629D/N629D yolk sac vessels and aorta consist of sinusoids without normal arborization. Isolate...

Reproductive technologies are widely used in cattle, although many are associated with high-embryonic mortality, especially during early gestation, when the yolk sac undergoes macroscopic changes in structure. We hypothesized that vasculogenesis and angiogenesis are affected, thereby affecting embryonic and placental differentiation. To test this, we studied yolk sac development and gene expres...
